Genetic diversity of Francisella tularensis in Poland with comments on MLVA genotyping and a proposition of a novel rapid v4-genotyping

We investigated genotypes of F. tularensis strains isolated in Poland in 1953-2013 and their genetic relationship to F. tularensis strains isolated in other countries using MLVA. Using PCR we examined mosquitos and ticks collected in Poland for the presence of F. tularensis. Our results revealed high genetic diversity of F. tularensis in Poland which might suggest that the bacterium is common in the environment. But arthropod bites are not a main source of infection because we do not confirm the presence of F. tularensis in ticks and mosquitos. We also proposed a practical assay called v4-genotyping that can be performed on the clinical and environmental samples directly. In addition, we discovered genetic differences among Schu S4 reference strains in various laboratories and showed that MLVA analysis should not be based on amplicon sizes only because point mutations occurring within the MLVA loci might not be reflected in an amplicon size.
